Summary
Real time quantitative PCR using TaqMan® probes. (1) In intact TaqMan® probes, reporter fluorescence is quenched due to FRET. (2) Probes and the complementary DNA strand are hybridized and reporter fluorescence is still quenched. (3) During PCR, the 5'-ends of the probe are degraded by the Taq polymerase. The fluorescent reporter is released.
